Rolker Gracie was born in Rio de Janeiro, Brazil, and is the fourth son of Helio Gracie, one of the founders of Gracie Jiu-Jitsu. He began training Brazilian Jiu-Jitsu at approximately six years old. While he did not have a major competitive career, his technical knowledge established him as a respected coach. In the 1990s, after his brother Rickson Gracie moved to the United States, Rolker became one of the head instructors at Gracie Humaita. Alongside his brother Royler Gracie, he helped build the academy into a top competitive team. After many instructors relocated abroad, Rolker remained in Rio de Janeiro as the main coach of the organization's headquarters.
